In August 2008, Shamim Dahod (CGS\u201976, CAS\u201978, MED\u201987) and her husband Ashraf gave $10.5M to BUSM to establish the Shamim and Ashraf Dahod Breast Cancer Research Center, as well as these programs and endowments.<o:p><\/o:p><\/p>\n<p class=\"xxxxxxmsonormal\" align=\"center\"><b><span>2022 Dahod International Scholar<\/span><\/b><o:p><\/o:p><\/p>\n<p class=\"xxxxxxmsonormal\"><b><span><img loading=\"lazy\" src=\"\/gms\/files\/2022\/03\/image012.jpg\" alt=\"\" width=\"108\" height=\"108\" class=\"size-full wp-image-85550 alignleft\" srcset=\"https:\/\/www.bumc.bu.edu\/gms\/files\/2022\/03\/image012.jpg 108w, https:\/\/www.bumc.bu.edu\/gms\/files\/2022\/03\/image012-100x100.jpg 100w\" sizes=\"(max-width: 108px) 100vw, 108px\" \/>Jingyi Zhao, PhD<\/span><\/b><span>, a postdoc in Dr. Mikel Garcia-Marcos\u2019 laboratory, will <\/span><span>characterize a small molecule inhibitor of a novel signaling mechanism that promotes breast cancer metastasis. He has identified a promising candidate molecule, named IGGi-11, that disrupts a protein signaling complex specifically present in metastatic cancer cells. He intends to further confirm the specificity of this molecule in preventing cancer cell invasiveness without overt cytotoxic effects in normal cells, and to work towards developing analog compounds with improved properties in preclinical breast cancer models.<\/span><o:p><\/o:p><\/p>\n<p class=\"xxxxxxmsonormal\" align=\"center\"><b><span>Dahod Pilot Grant Awardees<\/span><\/b><o:p><\/o:p><b><span>\u00a0<\/span><\/b><o:p><\/o:p><\/p>\n<p class=\"xxxxxxmsonormal\"><b><span><img loading=\"lazy\" src=\"\/gms\/files\/2022\/03\/image013.jpg\" alt=\"\" width=\"108\" height=\"108\" class=\"size-full wp-image-85551 alignleft\" srcset=\"https:\/\/www.bumc.bu.edu\/gms\/files\/2022\/03\/image013.jpg 108w, https:\/\/www.bumc.bu.edu\/gms\/files\/2022\/03\/image013-100x100.jpg 100w\" sizes=\"(max-width: 108px) 100vw, 108px\" \/>Daniel Dempsey, PhD, assistant professor of dermatology<\/span><\/b><span>, will collaborate with William Fairfield Warren Distinguished Professor of Biochemistry <b>Catherine Costello, PhD<\/b>, and Associate Professor of Pathology &amp; Laboratory Medicine and Ophthalmology <b>Nader Rahimi, PhD<\/b>, to develop new chemical biology tools to dissect the role of protein modifications in promoting breast cancer to precisely quantitate abnormally modified proteins and define a mechanistic purpose for these changes. Unraveling these mechanisms will identify new vulnerabilities that may be exploited with novel therapeutics to treat patients with breast cancer.<\/span><o:p><\/o:p><\/p>\n<p class=\"xxxxxxmsonormal\"><b><img loading=\"lazy\" src=\"\/gms\/files\/2022\/03\/image014.jpg\" alt=\"\" width=\"108\" height=\"108\" class=\"size-full wp-image-85552 alignleft\" srcset=\"https:\/\/www.bumc.bu.edu\/gms\/files\/2022\/03\/image014.jpg 108w, https:\/\/www.bumc.bu.edu\/gms\/files\/2022\/03\/image014-100x100.jpg 100w\" sizes=\"(max-width: 108px) 100vw, 108px\" \/>Hui Feng, MD, PhD, associate professor of pharmacology and medicine<\/b>, will study what nutrients metastatic triple-negative breast cancer (TNBC) cells use to spread to other organs and how they manage to survive in a harsh environment different from the mammary gland. Because genetically unstable tetraploid cells are a major contributor to breast cancer development, this work may suggest new therapeutic strategies to specifically impair DNA replication in whole genome-doubled breast cancer cells while sparing normal cells.<o:p><\/o:p><\/p>\n<p class=\"xxxxxxmsonormal\"><b><span><img loading=\"lazy\" src=\"\/gms\/files\/2022\/03\/image017.jpg\" alt=\"\" width=\"108\" height=\"108\" class=\"size-full wp-image-85556 alignleft\" srcset=\"https:\/\/www.bumc.bu.edu\/gms\/files\/2022\/03\/image017.jpg 108w, https:\/\/www.bumc.bu.edu\/gms\/files\/2022\/03\/image017-100x100.jpg 100w\" sizes=\"(max-width: 108px) 100vw, 108px\" \/>Alla Grishok, PhD, associate professor of biochemistry<\/span><\/b><span>,\u00a0studies the oncoprotein <\/span><span>DOT1L, which is essential for triple negative breast cancer progression. DOT1L, localized to the nucleus, activates many genes driving uncontrolled cell divisions.\u00a0The Grishok lab\u00a0will study how different parts of the large DOT1L protein work together to activate cancer-promoting genes. This mechanistic work may suggest new approaches to inhibit DOT1L, a promising drug target for cancer treatment.<\/span><o:p><\/o:p><\/p>\n<p class=\"xxxxxxmsonormal\"><b><span><img loading=\"lazy\" src=\"\/gms\/files\/2022\/03\/image018.jpg\" alt=\"\" width=\"108\" height=\"108\" class=\"size-full wp-image-85557 alignleft\" srcset=\"https:\/\/www.bumc.bu.edu\/gms\/files\/2022\/03\/image018.jpg 108w, https:\/\/www.bumc.bu.edu\/gms\/files\/2022\/03\/image018-100x100.jpg 100w\" sizes=\"(max-width: 108px) 100vw, 108px\" \/>Valentina Perissi, PhD, associate professor of biochemistry<\/span><\/b><span>, will study the molecular mechanism of PARP1 inhibition in human breast cancer cells. Poly-ADP ribosyltransferase inhibitors (PARPi) have emerged as promising\u00a0drugs\u00a0for the treatment of triple negative breast cancer and metastatic breast cancer.\u00a0Understanding how PARP activity is endogenously restricted in cells\u00a0will assist in determining the dosage of current PARPi, their efficacy in combination with other treatments, and the design of novel inhibitors.<\/span><o:p><\/o:p><\/p>\n<p class=\"xxxxxxmsonormal\"><b><span><img loading=\"lazy\" src=\"\/gms\/files\/2022\/03\/image019.jpg\" alt=\"\" width=\"108\" height=\"108\" class=\"size-full wp-image-85558 alignleft\" srcset=\"https:\/\/www.bumc.bu.edu\/gms\/files\/2022\/03\/image019.jpg 108w, https:\/\/www.bumc.bu.edu\/gms\/files\/2022\/03\/image019-100x100.jpg 100w\" sizes=\"(max-width: 108px) 100vw, 108px\" \/>Jessica Petrick MPH, PhD, assistant professor of medicine<\/span><\/b><span>, will use data and samples from the Black Women\u2019s Health Study to see if inflammation-related biomarkers are associated with an increased breast cancer risk in Black women, who have the highest breast cancer mortality rate of any racial or ethnic group in the U.S. Black Americans experience a disproportionately high burden of psychosocial distress due to individual and systemic racism. Experiences of racism are associated with increased inflammation in Black Americans, which may in turn influence risk of cancer and other chronic illnesses. <\/span><o:p><\/o:p><\/p>\n<p class=\"xxxxxxnih-ht10\"><b><span><img loading=\"lazy\" src=\"\/gms\/files\/2022\/03\/image020.jpg\" alt=\"\" width=\"108\" height=\"108\" class=\"size-full wp-image-85559 alignleft\" srcset=\"https:\/\/www.bumc.bu.edu\/gms\/files\/2022\/03\/image020.jpg 108w, https:\/\/www.bumc.bu.edu\/gms\/files\/2022\/03\/image020-100x100.jpg 100w\" sizes=\"(max-width: 108px) 100vw, 108px\" \/>Hiroshi Tokuo, MD, PhD<\/span><\/b><span>, <b>research assistant professor of physiology &amp; biophysics<\/b>,<span class=\"xxxxxxapple-converted-space\">\u00a0<\/span>will study how stromal cells communicate with breast cancer cells. Cancer cells are connected with immune cells through tunneling nanotubes, which support mitochondrial transfer. He will use <i>in vitro<\/i><span class=\"xxxxxxapple-converted-space\">\u00a0<\/span>and<span class=\"xxxxxxapple-converted-space\">\u00a0<\/span><i>in vivo<\/i><span class=\"xxxxxxapple-converted-space\">\u00a0<\/span>tumor models to see how these connections are established and single-cell RNA sequencing analysis to determine how mitochondrial transfer<span class=\"xxxxxxapple-converted-space\">\u00a0<\/span>metabolically empowers the cancer cells and depletes the immune cells.<\/span><o:p><\/o:p><\/p>\n<p class=\"xxxxxxmsonormal\"><b><span><img loading=\"lazy\" src=\"\/gms\/files\/2022\/03\/image021.jpg\" alt=\"\" width=\"108\" height=\"108\" class=\"size-full wp-image-85560 alignleft\" srcset=\"https:\/\/www.bumc.bu.edu\/gms\/files\/2022\/03\/image021.jpg 108w, https:\/\/www.bumc.bu.edu\/gms\/files\/2022\/03\/image021-100x100.jpg 100w\" sizes=\"(max-width: 108px) 100vw, 108px\" \/>Bob Varelas, PhD<\/span><\/b><span>, <b>associate professor of biochemistry, <\/b>will collaborate with\u00a0<b>Stefano Monti, PhD<\/b>,<b> <\/b>associate professor of medicine and biostatistics, to define and understand tumor-stromal signaling networks in triple negative breast cancer.
