They will assess data quality and identify the most effective tissue\u2011sampling approaches for creating accurate and representative views of each tumor\u2019s immune environment.<\/p>\n\n\n\n<div class=\"wp-block-image is-style-rounded\"><figure class=\"alignleft size-large\"><img loading=\"lazy\" width=\"143\" height=\"180\" src=\"\/camed\/files\/2026\/02\/thomas-Clarke.jpg\" alt=\"Portrait of Thomas Clarke in blue shirt standing in front of green shrub outside\" class=\"wp-image-141933\"\/><\/figure><\/div>\n\n\n\n<p><strong><a href=\"https:\/\/profiles.bu.edu\/Thomas.Clarke\">Thomas Clarke<\/a>, PhD, assistant professor of pathology &amp; laboratory medicine<\/strong>, and collaborators have discovered a previously uncharacterized zinc-finger protein that helps repair damage to DNA. In triple-negative breast cancer (TNBC), this protein is often reduced, and low levels are linked to poorer outcomes in advanced disease. Using spatial transcriptomics on human and murine tissue, they will map where MSCs reside, characterize the surrounding cellular microenvironment, and identify tumor derived signals that preserve MSC stemness and drive CAF differentiation for targeted intervention.<\/p>\n\n\n\n<div class=\"wp-block-image is-style-rounded\"><figure class=\"alignright size-large\"><img loading=\"lazy\" width=\"180\" height=\"179\" src=\"\/camed\/files\/2025\/11\/Ignaty-Leshchiner-PhD.jpg\" alt=\"headshot of Ignaty Leshchiner, PhD\" class=\"wp-image-139943\" srcset=\"https:\/\/www.bumc.bu.edu\/camed\/files\/2025\/11\/Ignaty-Leshchiner-PhD.jpg 180w, https:\/\/www.bumc.bu.edu\/camed\/files\/2025\/11\/Ignaty-Leshchiner-PhD-150x150.jpg 150w, https:\/\/www.bumc.bu.edu\/camed\/files\/2025\/11\/Ignaty-Leshchiner-PhD-100x100.jpg 100w\" sizes=\"(max-width: 180px) 100vw, 180px\" \/><\/figure><\/div>\n\n\n\n<p><strong><a href=\"https:\/\/profiles.bu.edu\/Ignaty.Leshchiner\">Ignaty Leshchiner<\/a>, PhD, assistant professor of medicine\/computational biomedicine<\/strong>, and his team previously&nbsp;identified that metastatic ER-positive breast cancers frequently develop resistance to targeted inhibitors and endocrine therapy through diverse, coexisting mechanisms, including convergent loss-of-function alterations in the chromatin modifier genes across independent resistant subclones, driving reduced ER signaling and multidrug resistance. The work aims at identifying biomarkers that predict responsiveness and guide the development of novel inhibitor strategies to improve therapeutic outcomes.<\/p>\n\n\n\n<div class=\"wp-block-image is-style-rounded\"><figure class=\"alignleft size-large\"><img loading=\"lazy\" width=\"159\" height=\"180\" src=\"\/camed\/files\/2015\/06\/Xaralabos-Varelas-3.jpg\" alt=\"Headshot of Bob Varelas\" class=\"wp-image-117074\"\/><\/figure><\/div>\n\n\n\n<p><strong><a href=\"https:\/\/profiles.bu.edu\/Xaralabos.Varelas\">Bob Varelas<\/a>, PhD, professor of biochemistry &amp; cell biology<\/strong>, will study how common environmental chemicals, including \u201cforever chemicals\u201d and hormone-disrupting compounds like BPA, may combine with normal aging changes in breast tissue to increase cancer risk.
