Xiaofeng’s research at BUCBM, under the leadership of Prof. Zaia, focuses on the biochemistry, especially structural determination of glycosaminoglycans that serve a multitude of important biological functions. He is primarily involved in the development of mass spectrometry-based platforms to gain biochemical insights of structurally polydisperse heparan sulfate and chondroitin sulfate. The platforms encompass GAG extraction and purification from a variety of biological sources, online LC separation, ionization manipulation, MS detection, MS/MS structure determination of different dissociation modes, and GAG labeling and chemical modification. With the established and continuously improving platforms, he investigates how heparan sulfate and chondroitin sulfate are expressed and exhibit structural differences among samples from sources of different disease states, development stages, and genetic backgrounds. He participates in a number of projects that are collaborative efforts between intramural researchers at BUMC and scholars around the world.
